چکیده
We have previously shown (Iles et al., 1980) that the metabolism of fructose may be followed in isolated rat livers by using 3*P n.m.r. spectroscopy. The introduction of the surfacecoil technique (Ackerman et al., 1980) makes it possible to study metabolism in the anaesthetized animal. A fed male Wistar rat was anaesthetized by intraperitoneal sodium pentobarbital, catheterized in the right jugular vein and secured vertically within a 7cm n.m.r. probe. A flat, three-turn IOmm-diameter radiofrequency coil was placed on the surface of the left lobe of the liver through an abdominal incision. To prevent evaporation, and to electrically insulate the coil, the liver surface was covered with thin plastic film. The probe was inserted into a wide-bore Oxford Instruments superconducting (4.3 T) magnet. Radiofrequency pulses were repeated every

Effect of graded hypoxia on the rat hepatic tissue oxygenation and energy metabolism monitored by near-infrared and 31P nuclear magnetic resonance spectroscopy.
Alteration in hepatic cellular adenosine triphosphate (ATP) levels has been shown to be a sensitive index for hypoxic damage. Hepatic ATP metabolism can be monitored by 31P nuclear magnetic resonance (NMR). Near-infrared spectroscopy (NIRS) can measure tissue oxyhemoglobin (HbO2), deoxyhemoglobin (Hb), and cytochrome oxidase (Cyt Ox), which reflect ATP production. In this study, hepatic oxygena...
متن کاملFructose-induced aberration of metabolism in familial gout identified by 31P magnetic resonance spectroscopy.
The hyperuricemia responsible for the development of gouty arthritis results from a wide range of environmental factors and underlying genetically determined aberrations of metabolism. 31P magnetic resonance spectroscopy studies of children with hereditary fructose intolerance revealed a readily detectable rise in phosphomonoesters with a marked fall in inorganic phosphate in their liver in viv...
متن کاملEffect of cyclosporine on hepatic energy status and on fructose metabolism after portacaval shunt in dog as monitored by phosphorus-31 nuclear magnetic resonance spectroscopy in vivo.
The effect of cyclosporin A on the hepatic energy status and intracellular pH of the liver and its response to a fructose challenge has been investigated using in vivo phosphorus-31 nuclear magnetic resonance spectroscopy in dogs. Three experimental groups were studied: (a) control dogs (n = 5), (b) dogs 4 days after the creation of an end-to-side portacaval shunt (n = 5), and (c) dogs 4 days a...
متن کاملHepatocyte anoxic injury is prevented by high concentration of fructose.
H UMAN livers can only be stored for a short time before transplantation, thus a better understanding of poteatial phannacologic and nutritional strategies that might protect the liver against anoxic damage may be imponant clinically. Fructose has been shown recently to prevent liver damage induced by Watnl ischemia as assessed by lactate dehydrogenase (LDH) release. I To determine whether fruc...
متن کامل31P and 13C nuclear magnetic resonance studies of metabolic pathways in Pasteurella multocida characterization of a new mannitol-producing metabolic pathway.
Glucose metabolism of Pasteurella multocida was examined in resting cells in vivo using 13C NMR spectroscopy, in cell-free extracts in vitro using 31P NMR spectroscopy and using enzyme assays. The NMR data indicate that glucose is converted by the Embden-Meyerhof and pentose phosphate pathways. The P. multocida fructose 6-phosphate phosphotransferase activity (the key enzyme of the Embden-Meyer...
